Kuno Wildlife Sanctuary was established in 1981 with an initial area of about {{convert|344.68|km2|sqmi|abbr=on}}.&amp;lt;ref name=Kabra2009/&amp;gt;&lt;br /&gt;
In the 1990s, it was selected as a possible site to implement the [[Asiatic Lion Reintroduction Project]], which aimed at establishing a second lion population in India.&amp;lt;ref name=Khudsar_al2008&amp;gt;{{cite journal |last1=Khudsar |first1=F. Between 1998 and 2003, about 1,650 inhabitants of 24 villages were resettled to sites outside the protected area.&amp;lt;ref name=Kabra2009/&amp;gt;&lt;br /&gt;
Most of the inhabitants were [[Saharia]] tribal people. An area of {{convert|924|km2|sqmi|abbr=on}} surrounding the wildlife sanctuary was added as buffer zone to human settlements.&amp;lt;ref name=Khudsar_al2008/&amp;gt;&lt;br /&gt;

The vegetation of the protected area includes &amp;#039;&amp;#039;[[Anogeissus]] pendula&amp;#039;&amp;#039; forest and scrub, &amp;#039;&amp;#039;[[Boswellia]]&amp;#039;&amp;#039; and &amp;#039;&amp;#039;[[Butea]]&amp;#039;&amp;#039; forest, dry savanna forest and grassland and tropical [[riverine forest]]. The dominant tree species are &amp;#039;&amp;#039;[[Acacia catechu]]&amp;#039;&amp;#039;, Salai &amp;#039;&amp;#039;[[Boswellia serrata]]&amp;#039;&amp;#039;, Tendu &amp;#039;&amp;#039;[[Diospyros melanoxylon]]&amp;#039;&amp;#039;, Palash &amp;#039;&amp;#039;[[Butea monosperma]]&amp;#039;&amp;#039;, Dhok &amp;#039;&amp;#039;[[Anogeissus latifolia]]&amp;#039;&amp;#039;, &amp;#039;&amp;#039;[[Acacia leucophloea]]&amp;#039;&amp;#039;, &amp;#039;&amp;#039;[[Ziziphus mauritiana]]&amp;#039;&amp;#039; and &amp;#039;&amp;#039;[[Ziziphus]] xylopyrus&amp;#039;&amp;#039;. Prominent shrub species include &amp;#039;&amp;#039;[[Grewia]] flavescens&amp;#039;&amp;#039;, &amp;#039;&amp;#039;[[Helicteres isora]]&amp;#039;&amp;#039;, &amp;#039;&amp;#039;[[Hopbush]] viscosa&amp;#039;&amp;#039;, &amp;#039;&amp;#039;[[Vitex negundo]]&amp;#039;&amp;#039;. Grass species include &amp;#039;&amp;#039;[[Heteropogon contortus]]&amp;#039;&amp;#039;, &amp;#039;&amp;#039;[[Apluda]] mutica&amp;#039;&amp;#039;, &amp;#039;&amp;#039;[[Aristida]] hystrix&amp;#039;&amp;#039;, &amp;#039;&amp;#039;[[Themeda quadrivalvis]]&amp;#039;&amp;#039;, &amp;#039;&amp;#039;[[Cenchrus ciliaris]]&amp;#039;&amp;#039; and &amp;#039;&amp;#039;[[Desmostachya bipinnata]]&amp;#039;&amp;#039;. &amp;#039;&amp;#039;[[Senna tora]]&amp;#039;&amp;#039; and &amp;#039;&amp;#039;[[Argemone mexicana]]&amp;#039;&amp;#039; are also common.&amp;lt;ref name=Bipin_al2003&amp;gt;{{cite book |authors=Bipin, C. The main predators occurring in the protected area are [[Indian leopard]], [[jungle cat]], [[sloth bear]], [[dhole]], [[Indian wolf]], [[golden jackal]], [[striped hyena]] and [[Bengal fox]]. Ungulates include [[chital]], [[Sambar deer]],  [[nilgai]], [[four-horned antelope]], [[chinkara]], [[blackbuck]] and [[wild boar]].&amp;lt;ref name=Khudsar_al2008/&amp;gt;&amp;lt;ref name=Bipin_al2003/&amp;gt; More than 1,900 [[feral cattle|feral]] [[zebu]] were estimated to occur in 2008, whereas density of wild ungulates was considered too low to sustain an introduced lion population at the time.&amp;lt;ref name=Khudsar_al2008/&amp;gt;&lt;br /&gt;

[[Honey badger]], [[Indian grey mongoose]], [[ruddy mongoose]], [[small Asian mongoose]], [[gray langur]], [[Indian crested porcupine]] and [[Indian hare]] have also been recorded. Reptiles present include [[mugger crocodile]], [[Gharial]], [[Bengal monitor]] and [[Indian softshell turtle]].&amp;lt;ref name=Bipin_al2003/&amp;gt;&lt;br /&gt;

A total of 129 bird species were sighted during a survey in spring 2007. [[Indian white-backed vulture]], [[long-billed vulture]], [[red-headed vulture]], [[Egyptian vulture]], [[crested serpent-eagle]], [[short-toed snake eagle]] [[Bonelli&amp;#039;s eagle]], [[white-eyed buzzard]], [[changeable hawk-eagle]], [[brown fish owl]] and [[spotted owlet]] are resident raptors.
